How does the reply link work in the Campaign Manager, is possible to have stats for if they click on a link (to page of website) in the mail?

Yes - when a user clicks on a reply option it is recorded in the database and you can see how many users clicked on an option and which one.
Ok, how do i put in the link i want to send them to, like now you have <a href="{reply:4}">, and i want to send them to "www.domain.com/page/index.htm"
No in such way you can't get stats. Built-in Campaign manager links are intended to receive some answer from the target - like "Would you like to participate in our contest? Answer Yes or No.". You custom links should be inserted with editor and you won't get any stats for them.
